Precautions when using fermenter
Nov 27, 2019| Fermenters are suitable for fermentation reactions between different strains and materials, including aerobic fermentation and anaerobic fermentation.The equipment in the use of a variety of bacteria and raw material ratio requirements are very strict.The ferment tank is made of advanced stainless steel and is welded to a cylinder, oval or disk end.Through the stirring device in the tank, the materials are mixed evenly, the mixing effect is good, and the fermentation reaction between the bacteria and the raw materials is promoted. The high-temperature sterilization can be realized, which conforms to the national GMP hygiene standard, and is widely used in food, medicine, chemical industry and other industries.
Fermentation tank is mainly composed of tanks, jacket, mixing system three system structure, setting up jacket or coil, gear, shaft sealing device, heat preservation, support, fermentation medium special sampling import and export, feeding valve and coolant temperature sensor interface, PH automatic control system, pressure sensor, air respirator, CIP interface and other components.Below we introduce the characteristics of the fermenter and the use of note.
I. characteristics of fermenter
(1) uniform distribution of reaction liquid: uniform gas-liquid-solid three-phase mixing and good mixing and dispersion of solution components are common requirements of bioreactors, because the distribution of flow, mixing and residence time are affected.For many intermittent or continuous aeration fermentations, the matrix and dissolved oxygen are dispersed as evenly as possible to ensure that the matrix concentration in the fermentation tank is between 0.1% and less than 1%, and the dissolved oxygen concentration is between 10% and 30%.This is conducive to the growth of aerobic cells and the formation of products.In addition, the formation of a stable foam layer at the fermentation tank level must be avoided to avoid the accumulation, destruction and even death of biological cells.There are also intermediate components, particularly starchy granular materials that are prone to precipitation and should be able to suspend and disperse.The gas-lift loop reactor can well meet these requirements.
(2) high dissolved oxygen rate and dissolved oxygen efficiency: the gas-lift reactor has high gas-holdup and gas-liquid contact interface, with high mass transfer rate and dissolved oxygen efficiency. The volume dissolved oxygen efficiency is usually higher than that of the mechanical stirring tank, KLD can reach 2000 hours, and the dissolved oxygen energy consumption is low.
(3) low cutting force and low damage to biological cells caused by fermentation tank: since there is no mechanical stirring impeller in the air-lift reactor, the shear damage of cells is reduced to a low level, which is especially suitable for the culture of plant cells and tissues.
Ii. Matters needing attention when using fermenter
1. This system must be used when all equipment in the fermentation tank is working properly.
2. When sterilizing the filter, the vapor pressure through the air filter shall not exceed 0.17 miles.Otherwise, the filter element will be damaged and the filter capacity will be lost.
3. During the fermentation process, the pressure in the tank should not exceed 0.17mpa.
4. In the actual consumption process, when the jacket is preheated by steam, the inlet steam pressure must be controlled within the working pressure range of the equipment (no more than 0.2mpa), otherwise the fermenter will be damaged.
5. During emptying and actual emptying, the remaining water must be discharged into the sheath of the fermentation tank, otherwise, the inner cylinder of the fermentation tank may be compressed, causing equipment damage.In addition, too much condensed water will lead to dilution of the medium, which cannot meet the process requirements.
6. It is strictly prohibited to use the negative pressure in the fermentation tank during cleaning the empty tank and cooling process after the actual dismantling, so as to avoid pollution and even damage to the equipment.
7. During the fermentation process, the pressure of the fermentation tank should be kept between 0.03-0.05mpa to avoid contamination.
8. During each operation, the pressure in the trachea must be higher than that in the fermentation tank, otherwise the liquid in the fermentation tank will return to the filter and block the filter element or make the filter fail
